在新加坡，一家籠罩著許多秘密和神秘氣息的古老店屋引起了銀行家志玲的注意。她從香港返回新加坡，為的就是領取遺產。在領取遺產的當兒，她漸漸走進曾祖母即神秘又黑暗的世界，並無意中揭發了這間設立於新加坡40年代初的妓院的過去。而這座歷史性建築的命運，將落在志玲的手中。她必須找尋自己的根源，瞭解家族的歷史。《最後一位夫人》講述的是志玲和曾祖母鳳嵐的迷人故事，兩個來自不同世界和時代的兩人看似陌生，可是卻有著微妙的關係。
